Is Glucosamine or MSM the Right First Move?
Sadie was an 8-year-old Border Collie, 42 pounds, the kind of dog who used to clear a coffee table jump without thinking about it. Her owner noticed the change on a Tuesday in early spring: Sadie stood up from her afternoon nap on the hardwood floor, held her weight oddly on the left hind leg for three or four steps, and then walked it off. It happened again that weekend, after a longer hike. Nothing dramatic — no yelp, no limp that lasted more than a minute — but her owner had raised two other dogs into their teens and knew what the first quiet season of joint trouble looks like. She came into my exam room with a question I hear almost weekly in one form or another: glucosamine, MSM, or both? Not every study is a wash. A Randomised double-blind, positive-controlled trial to assess the efficacy of glucosamine/chondroitin sulfate for the treatment of dogs with osteoarthritis: randomised double-blind, positive-controlled trial comparing glucosamine/chondroitin sulfate against carprofen in osteoarthritic dogs found statistically significant improvements in pain, weight-bearing, and severity scores by day 70 — though the response came on slower than the drug did. A more recent trial in dogs with hip osteoarthritis, the Study of the Effectiveness of Glucosamine and Chondroitin Sulfate, Marine-Based Fatty Acid Compounds, and Carprofen for the Treatment of Dogs with Hip Osteoarthritis: Study of the Effectiveness of Glucosamine and Chondroitin Sulfate, Marine-Based Fatty Acid Compounds, and Carprofen published in PMC, found the opposite for glucosamine specifically: increases in peak vertical force (a gait-based measure of how much weight a dog puts on a limb) were not significantly greater than placebo at four and six weeks, while the marine fatty acid compounds and carprofen groups did significantly improve. Two well-controlled trials, two different answers — that inconsistency is the honest shape of the glucosamine evidence base.
Where MSM Fits — and Where the Evidence Runs Thin
MSM supplies bioavailable sulfur, which the body uses in collagen cross-linking and connective tissue synthesis, and it’s often framed as reducing the inflammatory signaling that drives joint discomfort. The mechanism is plausible and the sulfur-collagen connection is real chemistry, not marketing fiction. What’s thinner is dog-specific trial data. That’s a human study, not a canine one, and I don’t extrapolate human dosing or outcomes onto dogs — different metabolism, different trial population. The Case for Combining Them, and the Counterargument I Take Seriously
Here’s where I land, and I’ll name the pushback first because it’s a fair one: if the canine glucosamine data is genuinely mixed and the MSM data is mostly borrowed from human trials, why recommend either? The counterargument is that you’re spending money and pill burden on a combination with no dog-specific proof of additive benefit.
I still recommend a combination product for most dogs starting mobility support, and here’s the reasoning. The mechanisms are complementary rather than redundant — one is a cartilage-matrix substrate, the other is a sulfur donor and proposed anti-inflammatory — so there’s biological plausibility for them working on different parts of the same problem rather than competing for the same effect. If you see improvement by that point, some clinicians and labels support tapering to an every-other-day maintenance dose — but that’s a conversation to have with your own vet given your dog’s weight, kidney and liver status, and any other medications on board, not a default plan.

So Which One Should You Buy First?
